Biological Activity
Purmorphamine is a 2,6,9-trisubstituted purine compound that selectively induces osteogenesis in multipotent mesenchymal progenitor cells. The small molecule Purmorphamine has been found to activate Hedgehog (Hh) signaling pathway by directly targeting Smoothened (Smo), resulting in the up- and down-regulation of its downstream target genes, including Gli1 and Patched. The EC50 of purmorphamine for differentiation of C3H10T1/2 cells based on alkaline phosphatase (ALP) expression is 1 µM. This compound may ultimately be useful for the treatment of bone-related disease and neurodegenerative disease [1-3]. Purmorphamine can replace sonic hedgehog (SHH) in the entire process of ventral spinal progenitor specification and motor neuron differentiation with a similar efficiency [4].
