ZYMUPHEN TF-bearing Microparticles' Procoagulant（MP）-ACTIVITY（携带组织因子微粒促凝活性试剂盒）

Functional assay for the measurement of procoagulant activity of microparticles exposing Tissue Factor (TF) in human plasma, in vitro, using automated or manual method.

METHOD :
ZYMUPHEN MP-TF kit is a bio-immunoassay for the determination of microparticles exposing Tissue Factor (MP-TF), in human plasma or in purified milieu.
CAUTION: An automated microplate washer is required to use the kit.

APPLICATIONS :
Assay of MP-TF activity in some clinical contexts such as cancer, inflammation, diabetes, acute coronary syndrome or atherosclerosis.
Assay of MP-TF activity in cell culture supernatants.

TESTED SAMPLES :
- Human citrated, or CTAD-anticoagulated plasma (MP-specific specimen
collection and handling : refer § specimen collection, handling and storage)
- Cell culture supernatants
 

PRINCIPE :
In a first step, the AE-MP-TF solution and the sample are introduced into the wells of the microplate coated with a murine monoclonal antibody, specific for human Tissue Factor (TF, extracellular domain), and which does not interfere with TF activity. MP-TF present in the sample bind to the solid phase through an epitope localized in the extracellular domain of TF. Following an overnight incubation and an automated washing step, wash solution is immediately introduced into the wells. Then, Factor VIIa (R1) and Factor X (R2) are added. The TF-FVIIa complex forms and subsequently activates Factor X into activated Factor X (FXa), on the surface of the anionic phospholipids present in the microparticle, and in presence of calcium ions. The MP-TF concentration is the limiting
factor. In a third step, a specific substrate for FXa is introduced and reacts with FXa and produces a yellow color. The absorbance, recorded at 405nm on a spectrophotometer, is directly proportional to the amount of MP-TF present in the sample.
